摘要
目的:研究抑癌基因 wtp53在不同发育时期 Wistar 大鼠小肠上皮细胞的表达、分布特征以及与细胞增殖分化的关系,探讨 wtp53基因在小肠创伤修复时可能发挥的生物学作用。方法:利用 wtp53单克隆抗体,以超敏的 SP 免疫组织化学方法,观察 wtp53基因在胚胎、新生期各时间段 Wistar 大鼠小肠上皮细胞的阳性表达与分布特征。同时以细胞增殖活性的标志 PCNA 表达为参照,对不同时期细胞的增殖分化状况作对比性研究。结果:在胚胎第17d(E17d)、E19d、新生期第1(P1)d,小肠上皮细胞 wtp53基因的阳性表达范围较广,染色强度较深。P2d和 wtp53基因表达不仅范围扩大且染色强度明显增强;到幼鼠期第28d,wtp53基因在小肠上皮细胞的表达为阴性;从胚胎至 P2d,PCNA 的表达与 wtp53类似,而幼鼠期第28d,PCNA 阳性细胞仅局限于小肠绒毛隐窝。结论:在小肠发育的早期,上皮细胞的快速增殖分化期伴随着 wtp53基因的高度表达,可能对遗传物质 DNA的完整性起到监视和保护作用。
Objective:To investigate the relationship between the expression of the tumor-suppressor gene wtp53 and cellular proliferation and differentiation of intestinal epithelial cells in different developmental stages in rats, and to explore its biological role in repair of intestinal epithelium after injury.Methods:Specimens of tne intestine were obtained from rats in different developmental stages.Immunohisto-chemical techniques were used to detect the expressions of wtp53 and PCNA.Results:The protein expression of wtp53 was widely spread and strong in in- testinal epithelial cells in embryos of 17d,19d and postnatal 1d.Its expression appeared in larger area and stronger on postnatal 2d,whereas it become negative on postnatal 28d.The expression of PCNA was similar to wtp53 dur- ing the same periods.Though the expression of wtp53 on postnatal 28d was negative,the expression of PCNA could still he detected in intestinal crypts.Conclusions;The results indicate that the expression of wtp53 is parallel with PCNA at the early developmental stages of intestinal epithelial cells,wich may play roles in monitoring and protecting the integrality of DNA.
