中枢和外周神经损伤后胞体区看家基因表达的变化 被引量:1

摘要 目的:观察并比较几种常用的看家基因,包括β-actin,GAPDH,18s RNA,cyclophilin A 等在中枢和外周神经损伤后胞体分布区神经组织中表达的变化。方法:Wistar 雄性大鼠随机分为右侧坐骨神经夹伤,右侧坐骨神经结扎和胸段脊髓半横断三组。手术后7d,用相对定量 RT-PCR 的方法检测几种看家基因在受损神经元胞体分布区神经组织中的表达水平。结果:GAPDH 和 Cyclophilin A 的表达在不同模型中的变化幅度较小,损伤与对照侧比较差异均无显著性。β-actin 和18sRNA 的表达在不同模型中变化不同,在脊髓损伤和坐骨神经结扎后表达的变化有统计学意义。结论:本研究结果为研究神经系统损伤后相关基因表达变化时内参照基因的选择提供参考,同时也为进一步认识看家基因更加广泛的功能提供依据。 Objective;To investigate the expression changes in several housekeeping genes,including β-actin, GAPDH,18s RNA and cyclophilin A,in the neuronal somas after central nervous system (CNS) and peripheral nerve system (PNS) injury.Methods;Male Wistar rats were randomly divided into three groups;right sciatic nerve crush injury,right sciatic nerve ligation and hemisection of thoracic spinal cord.On the 7th day after injury, the expression levels of several housekeeping genes in neuronal somas associated with injured and control nerves were determined by semi-quantitative RT-PCR method.Results;The mRNA levels of GAPDH and Cycophilin A were not markedly changed in different nerve injury models,but β-actin and 18sRNA mRNA expressions were sig- nificantly different in neuronal somas after spinal cord injury as well as sciatic nerve ligation.Conclusion;This study provided a reference for selecting housekeeping genes in the observation of gene expression related to nerve injury,and the basis to explore the potential functions of housekeeping genes.
