摘要
目的:研究基质金属蛋白酶-2(MMP-2),基质金属蛋白酶-9(MMP-9)及其抑制因子(TIMP-2)在增生性瘢痕和正常皮肤中的表达特征。方法:用免疫组化技术检测这三种蛋白酶在12例增生性瘢痕和6例正常皮肤中的表达变化规律。结果:在正常皮肤中,MMP-2、MMP-9和TIMP-2的阳性率分别为(2.13±2.36)%、(16.25±18.72)%、(25.38±12.47)%。在增生性瘢痕中,三种蛋白的阳性细胞率明显升高(P<0.01),其中MMP-2和MMP-9主要定位于表皮细胞和成纤维细胞上,含有TIMP-2蛋白的阳性细胞也主要为表皮细胞和成纤维细胞。结论:MMP-2、MMP-9和TIMP-2可能对皮肤的发生、结构功能的维持以及创伤后修复起重要作用。
Objective:To explore the effect of matrix metalloproteinases (MMP-2,MMP-9) and its inhibitor (TIMP-2) on the formation of hypertrophic scars by investigating the localization and expression of these proteins in normal skin and hypertrophic scars.Methods:The expression intensity and distribution of MMP-2,MMP-9 and TIMP-2 were essessed with immunohistochemistry and pathological methods in 12 hypertrophic scars and 6 speci- mens of normal skin.Results:In normal skin,the positive rates of MMP-2,MMP-9 and TIMP-2 in cells were (2. 13±2.36)%,(16.25±18.72)% and(25.38±12.47)%,respectively.In hypertrophic scars,the positive rates of these proteins in cells were significantly increased compared with normal skin (P<0.01).The protein particles of MMP-2 and MMP-9 were principally distributed in cellular membrane of epidermal cells and fibroblasts.Positive signals of TIMP-2 were also distinctly located in epidermal cells and fibroblasts.Conclusion:MMPs and TIMPs might be involved in the development of the skin in embryonic stage,also in the cutaneous structure,as well as function maintenance and wound healing.
