卡巴胆碱对LPS刺激外周血白细胞释放TNFα的影响

Effect of carbachol on TNFαrelease from peripheral blood leukocytes stimulated by lipopolysaccharide

目的：观察拟胆碱药物卡巴胆碱对LPS刺激正常人和大鼠外周血白细胞释放TNFα的影响。方法：在LPS刺激的稀释全血或单个核细胞中加入卡巴胆碱孵育,用ELISA方法测定培养细胞上清液中TNFa的浓度,用免疫组化方法显示单个核细胞TNF表达量。结果：LPS刺激外周血白细胞释放TNFα,并呈时间依赖关系。LPS诱导单个核细胞TNFα表达增加,而卡巴胆碱抑制LPS的这些效应。结论：卡巴胆碱具有潜在的抗炎效应。

Objective:To investigate the effect of cholinergic drug carbachol on the release of TNFαfrom periph- eral blood leukocytes stimulated by LPS in human and rats.Methods:carbachol was added to the diluted peripheral blood or to mononuclear cells,to which LPS had been added,and then they were incubated for sertain period of time.The supernatants of culturs were harvested and assayed for TNFαby ELISA method.TNFαexpression in mononuclear cells was assessed by immunohistochemistry method.Results:LPS stimulated TNFαrelease from pe- ripheral blood leukocytes in a time-dependent pattern,and TNFαexpression in mononuclear cells was also en- hanced by LPS challenge.Both events could be abated by carbachol.Conclusion:The results suggested that carba- chol might possess a potential antiinflammatory effect.
