癫痫模型大鼠海马内fos基因表达及电子显微镜观察

摘要 [目的 ]观察癫痫模型大鼠海马的c fos基因表达 ,并探讨癫痫的发病机制 .[方法 ]采用fos蛋白免疫细胞化学方法和电子显微镜 ,观察注射戊四氮后 30min ,1,3,6h时的海马内fos蛋白阳性细胞的分布情况 .[结果 ]腹腔注射戊四氮后 30min时 ,海马内开始出现fos蛋白阳性细胞 ,1~ 2h时达到高峰 ,3h时开始逐渐减少 ,6h时基本消失 .[结论 ]海马在癫痫发作时反应强烈 ,同时有可复性的脑组织损伤 ,提示海马可能是癫痫发作的主要启动部位 . OBJECTIVE?To observe the expressions of c-fos in the hippocampus, to compare the fos positive cells at different time after seizures, and to study the mechanism of epilepsy.?METHODS?Using immunohistochemistry and electric microscope, it was studied that th e hi ppocampal tissues of rats after injecting the pentylenetetrazol at 30 minutres, one hour, three hours and six hours.?RESULTS?At 30 minutres a fter the abdominal injection of pentulenetetrazol, the fos positive cells we re begun to be found in the hippocampus and an hour later, the c-fos expression s reached the summit. After three hours, the num ber of fos positive cells were begun to decrease and six hours later, they were almost disappeared.?CO NCLUSION?A strong reaction had taken place when the epilepsy came o n. At the same time, there had some recoverable injuries in brain tissues. These indicate that the main beginning part of epilepsy in brain is the hippocampus.
