生物钟紊乱对糖尿病小鼠脏器损伤的影响 被引量:7

摘要 目的观察生物钟紊乱对链脲佐菌素(STZ)诱导的糖尿病小鼠肝、肾功能等的影响,并探讨其可能的机制。方法 30只昆明种小鼠随机分为对照组、糖尿病组和生物钟紊乱的糖尿病组。糖尿病组为禁食12h后腹腔注射STZ建立,生物钟紊乱的糖尿病组则同时改变光照时间,正常对照组仅注射等量生理盐水。测定各组小鼠24h尿蛋白定量、空腹血糖、血肌酐、血尿素氮、血清谷丙转氨酶、谷草转氨酶的含量、肝重系数、肾重系数。结果①与对照组相比,糖尿病组和生物钟紊乱的糖尿病组血糖浓度、肾重/体重系数(肾重系数)、肝重/体重系数(肝重系数)、24h尿蛋白定量、血肌酐、血清谷丙转氨酶、谷草转氨酶的含量均有不同程度的增加(P<0.01)。②与糖尿病组相比,生物钟紊乱的糖尿病组血糖浓度、肾重系数、肝重系数、24h尿蛋白定量、血肌酐、血清谷丙转氨酶、谷草转氨酶的含量均有不同程度的增加(P<0.01)。结论生物钟紊乱可以加速糖尿病肾病、糖尿病肝病的进程。 Objective To observe the effect of chronobiology disorder on kidney and liver injury in streptozotocin(STZ)-induced diabetic mice and to investigate the potential pathogenetic mechanisms.Methods Thirty KM mice were randomly divided into normal control group,diabetic model group,and chronobiology disorder model group(n=10 in each group).Diabetes was induced by a single peritoneal injection of streptozotocin after 12-hour fasting.Chronobiology disorder was induced by changing illumination rhythm in diabetic mice for 7 days.Mice in normal control group were injected with normal saline of the same volume.Then,the ratio of kidney weight and liver weight to body weight was calculated;and the 24 h urinary albumin,the fasting blood glucose(FBG),blood urea nitrogen(BUN),serum creatinine(Scr),alanine aminotransferase(ALT) and aspartate aminotransferase(AST) were measured.Results ① Compared with those in control group,the ratio of kidney weight and liver weight to body weight,the fasting blood glucose,BUN,Scr,the 24h urinary albumin,ALT and AST significantly increased in diabetic model group and chronobiology disorder model group(P<0.01).② In contrast to the mice in diabetic model group,the ratio of kidney weight and liver weight to body weight,the fasting blood glucose,BUN,Scr,the 24h urinary albumin,ALT and AST significantly increased to various extent in chronobiology disorder model mice(P<0.01).Conclusion These results indicate that chronobiology disorders may accelerate the development of kidney and liver injury in diabetic mice.
