摘要
纤维素酶应用广泛,但因受到酶活低、成本高等诸多因素的制约,纤维素酶的工业化生产和应用受到限制。应用基因工程技术来获得高活性、高产量的纤维素酶资源越来越受到人们的重视。本文对纤维素酶分子结构、基因的克隆、原核与真核表达进展进行了综述,并提出构建纤维素酶酵母表达系统、木霉表达系统以及多个纤维素酶基因共表达系统是未来纤维素酶基因工程的发展趋势。
Although cellulase is widely applied,large-scale industrial production of cellulase has been restricted by many factors,such as low enzyme activity and high cost.Increasing attention has been paid on genetic engineering technology,by which cellulase resources with high activity and high production are hopeful to be obtained.Molecular structure,gene cloning,prokaryotic and eukaryotic expression of cellulase genes are reviewed in this article.It is suggested that constructing of yeast expression system and Trichoderma expression system,as well as co-expression system of multiple cellulase genes would become the main stream of cellulase gene engineering.
