摘要
目的观察酪酪肽(PYY)对体外培养的人胰腺癌Miapaca-2细胞的生长抑制作用及Y受体在胰腺癌细胞的表达。方法不同浓度的PYY分别作用于体外常规培养Miapaca-2细胞,采用MTT比色法检测细胞增殖情况,光镜观察细胞形态变化。RT-PCR法检测胰腺癌细胞Y受体亚型Y1、Y2、Y5 mRNA表达。结果 PYY对Miapaca-2细胞生长有明显的抑制作用,呈剂量依赖性,与对照组相比差异有统计学意义(P<0.01);胰腺癌Miapaca-2细胞表达Y1、Y2和Y5受体,Y2受体表达较强。结论 PYY对体外培养的胰腺癌细胞有明显的抑制作用,胰腺癌Miapaca-2细胞主要表达Y2受体。
Objective To observe the effect of peptide YY(PYY) in inhibiting cell growth and detect Y receptors expression of human pancreatic carcinoma Miapaca-2 cell line.Methods The cell line of Miapaca-2 human pancreatic carcinoma was treated with PYY of different concentrations for 24,48 and 72 h,respectively.MTT assay was used to evaluate the cytotoxic effects of PYY on Miapaca-2 cells.Cellular morphous was observed with inverted microscope.The mRNA expressions of Y1,Y2 and Y5 receptor subtypes were detected with RT-PCR.Results Y1,Y2 and Y5 receptor subtypes were detected in human pancreatic cancer cell line Miapaca-2.PYY of different concentrations had a cytotoxic effect on Miapaca-2 cell line in obvious dose-and time-dependent manners.Conclusion PYY can inhibit Miapaca-2 cell growth in dose-and time-dependent manners.Y1,Y2 and Y5 receptor subtypes can be detected in human pancreatic cancer cell line Miapaca-2.
