小鼠Deaf1基因变构体全长cDNA的克隆

摘要 目的克隆小鼠Deaf1基因变构体全长cDNA序列。方法利用RT-PCR技术从非肥胖糖尿病(non-obesediabetic,NOD)小鼠胰淋巴结总RNA中克隆Deaf1变构体全长cDNA,并做多重序列比对。结果从NOD小鼠胰淋巴结中克隆得到了多个Deaf1变构体的全长cDNA序列,序列分析结果表明,部分Deaf1变构体缺失了重要功能结构域。结论 Deaf1在NOD小鼠胰淋巴结中存在不同的变构体,这些变构体的产生可能会影响Deaf1作为转录因子的功能并与1型糖尿病的发生密切相关。
