正交设计优选龙胆中龙胆苦苷的提取工艺

Optimization on extraction process of gentiopicroside by orthogonal design

目的优选龙胆中龙胆苦苷的最佳提取工艺。方法采用高效液相色谱法测定龙胆中龙胆苦苷的含量,以乙醇浓度、提取时间、料液比为考察因素,以龙胆苦苷提取率为考察指标,采用L9(34)正交设计优选最佳提取工艺条件。结果最佳提取工艺为提取溶剂70%乙醇,提取时间1.5 h,料液比1∶60。结论本法工艺简单、经济、提取率高,可为中药龙胆的开发利用提供参考。

Objective To optimize the extraction process of gentiopicroside from Gentiana scabra Bge..Methods A HPLC method was applied to determine the content of gentiopicroside.The best extraction technology was optimized by L9(34) orthogonal test with the concentration of ethanol,extraction time and material-liquid ratio as factors for solvent extraction process and the extraction rate of gentiopicroside was chosen as index.Results The optimal extraction process was as follows:extraction solvent of 70% ethanol,extracting time of 1.5 h and the material-liquid ratio of 1∶60(w/v).Conclusion The process is simple and economical with high extraction rate and can provide parameters for exploitation and utilization of Gentiana scabra Bge..
