摘要
目的研究抑制翻译过程对紫杉醇(PTX)诱导细胞凋亡的影响。方法体外培养人非小细胞肺癌A549;采用PTX单独或与翻译抑制剂cycloheximide联合处理细胞,采用流式细胞术分析细胞凋亡。结果2.5、5、10和20 nmol/L PTX作用24 h,细胞凋亡百分率分别为(16.70±2.08)%、(28.08±3.15)%、(17.12±2.57)%和(11.43±2.08)%;30 nmol/L cycloheximide使10和20 nmol/L PTX诱导凋亡效应分别增加了69.01%和165.62%。结论翻译抑制剂可增加10和20 nmol/L PTX诱导细胞凋亡效应,而对低浓度PTX诱导细胞凋亡无影响。
Objective To study the effect of inhibiting translating process to PTX on apoptosis.Methods Non-small cell cancer A549 was cultured in vitro,PTX or combination of PTX and cycloheximide were used to treat the cells,cell apoptosis was analyzed by flow cytometry.Results After 24 hours′ treating with the dose of PTX 2.5,5,10 and 20 nmol/L,cell apoptosis rates were(16.70±2.08)%,(8.08±3.15)2%,(17.12±2.57)% and(11.43±2.08)% respectively.30 nmol/L cycloheximide enhanced effect of apoptosis of 10 and 20 nmol/L PTX to 69.01% and 165.62%.Conclusion Cycloheximide could enhance effect of cell apoptosis of 10 and 20 nmol/L PTX and had no effect on cell apoptosis of PTX with low concentration.
