甲状腺滤泡癌PTEN和缺氧诱导因子1α的表达及其相关性 被引量:4

摘要 目的通过检测甲状腺滤泡癌组织中PTEN、缺氧诱导因子1α(HIF-1α)的表达,探讨PTEN和HIF-1α异常表达的相关性。方法采用免疫组织化学PV6000两步法检测甲状腺滤泡癌,甲状腺滤泡性腺瘤及结节性甲状腺肿标本中PTEN、HIF-1α的表达。结果 PTEN蛋白在结节性甲状腺肿、甲状腺滤泡性腺瘤中的阳性率分别为100%和80%,均显著高于甲状腺滤泡癌组织中的25%。HIF-1α在甲状腺滤泡癌中(60%)的表达明显高于甲状腺滤泡性腺瘤(30%)及结节性甲状腺肿(0)。结论甲状腺滤泡癌中PTEN表达缺失而HIF-1α表达明显增强,推测PTEN表达的缺失促进了HIF-1α表达,最终将促使甲状腺滤泡癌的发生、发展。 Objective To investigate the expression of PTEN and HIF-1α in human follicular thyroid carcinoma(FTC),and explore the correlation between them.Methods Expression of PTEN and HIF-1α in FTC,thyroid adenoma and goiter were detected by immunohistochemical PV6000 method.Results The positive rate of PTEN in nodular goiter,thyroid adenoma and FTC were 100%,80% and 25% .The positive rate of HIF-1α were 0,30% and 60% .The difference between them were of statistical significance.Conclusions Loss expression of PTEN and hyperexpression of HIF-1α play an important role in development of human follicular thyroid carcinoma.The loss of expression of PTEN may be promote the expression of HIF-1α.
