摘要
目的:探讨人胸腺组织胸腺基质淋巴细胞生成素(TSLP)表达水平与外周血CD4+CD25+Foxp3+Treg细胞的相关性。方法:取23例先天性心脏病患者,分离外周血单个核细胞后表面染色及破膜胞内染色后,流式细胞仪检测CD4+CD25+Foxp3+Treg,同时取对应患者切除的胸腺组织,免疫组织化学法检测其TSLP的表达水平,统计分析其差异性,Logistic回归分析二者相关性。结果:23例先心病患者CD4+CD25+Foxp3+Treg/CD4+T细胞比率为(5.73±0.56)%,胸腺Hassall小体TSLP表达水平为(18.87±3.06)个,Logistic回归分析表明二者呈正线性相关性。结论:人外周血CD4+CD25+Foxp3+Treg细胞的水平与胸腺组织TSLP的表达密切相关,外周血Treg细胞数量的增加可能依赖于更高水平TSLP的表达。
Objective To investigate the correlation between the expression level of thymic stromal lymphopoietin(TSLP) of human thymus tissue and the CD4+CD25+Foxp3+ Treg cells in peripheral blood mononuclear cells.Methods The CD4+CD25+Foxp3+Treg cell was tested by flow cytometry from peripheral blood mononuclear cell in 23 patients with congenital heart disease.The expression of TSLP in thymus gland was determined with immunohistochemical staining.The correlation between TSLP expression and CD4+CD25+Foxp3+Treg cell was analyzed by logistic regression.Results The ratio of CD4+CD25+Foxp3+Treg / CD4+T cell was(5.73 ± 0.56)% in 23 patients with congenital heart disease,and expression of TSLP in thymus Hassall’s corpuscle was 18.87±3.06,the logistic regression analysis demonstrated that the expression of TSLP and the phenotype of Treg cells was linear correlated.Conclusion The expression level of TSLP in human thymus is related to the phenotype of the CD4+CD25+Foxp3+Treg cells,and the increasing quantity of Treg cells in peripheral blood may depend on the higher level of TSLP expression.
