慢性阻塞性肺疾病大鼠造模不同时期肺组织及血中血管性血友病因子与一氧化氮含量的动态对比观察 被引量:1

摘要 研究慢性阻塞性肺疾病大鼠造模形成过程中不同时期大鼠血浆及肺组织匀浆中血管性血友病因子(VWF)与一氧化氮(NO)的动态变化。清洁级健康雄性Wistar大鼠48只,采用随机数字表法分为空白组24只,COPD模型组24只(COPD 1周组、COPD 2周组、COPD 4周组)。用气管内注入脂多糖(LPS)及烟熏方法制作慢性阻塞性肺疾病的模型。分别于第1、2、4周检测大鼠血浆及肺组织匀浆中VWF与NO的含量。实验组与空白组血浆和肺组织匀浆VWF、NO含量相对比均有统计学意义(P<0.05),其中,COPD 4周组和空白组对比具有显著差异(P<0.01);实验组中第1、2、4周相互之间比较存在统计学差异(P<0.05),其中,COPD 4周组与COPD1周组、COPD2周组相比较具有显著差异(P<0.01)。在慢性阻塞性肺疾病形成早期即出现血管内皮的损伤,并且随着COPD疾病的进展,VWF释放入血,血浆中VWF的含量增加,NO浓度则逐渐减低,说明血管内皮受损程度成逐渐加重的趋势。 To research VWF and NO's content changes in the plasma and the lung homogenate in different periods of COPD rat model.Choose 48 SPF male Wistar rats were choosen using random number table method,they were divided them into 2 groups:the blank group and COPD model group(COPD 1 week group,COPD 2 weeks group,COPD 4 weeks group).Blank group and COPD model group both has 24 Wistar rats.by gas supply tube into lipopolysaccharide(LPS) and smoked ways the chronic obstructive pulmonary disease model was made.VWF and NO's content of rats in the plasma and the lung homogenate were tested in 1,2,4 weeks.The content of VWF and NO in plasma and lung homogenate between experimental group and blank group were relatively statistical significance(P<0.05).Among them,COPD 4 weeks group and the blank group's contrast had significant differences(P<0.01);The contrast between Group 1,2,4 weeks had statistical differences(P<0.05).The compare between COPD 4 weeks group and COPD 1,2 weeks group also had significant differences(P<0.01);In early phase of chronic obstructive pulmonary disease,endothelial injury has appeared,and with the progress of the disease,VWF releases into the blood,the content of VWF increases,but the concentration of NO gradually decreases,shows that the extent of endothelial damage has increasing trend.
