高效黄瓜愈伤组织诱导培养的研究 被引量:1

摘要 以津研四号黄瓜种子为实验材料,对种子进行了无菌水浸泡及0.1%HgCl2消毒处理,探究了无菌实生苗培养的最佳条件;以无菌实生苗为外植体来源,研究了外植体类型及不同激素配比对黄瓜组织培养的影响。结果表明:种子用无菌水浸泡12 h,0.1%HgCl2消毒7 min,最适宜于无菌实生苗的培养;子叶、真叶、下胚轴三种外植体中,子叶更适于黄瓜愈伤组织的诱导培养;子叶愈伤组织诱导的最佳培养基为MS+0.8 mg/L 6-BA+0.10 mg/L IAA。 'Jingyan' NO.4 cucumber(Cucumis Sativus L.) was used as materials in the experiment.To investigate the best conditions for obtaining sterile seedlings,seeds were soaked in sterile water and disinfected with 0.1% HgCl2.Aseptic seedlings were used as explant source,the influence of different types of explant and hormone combination on callus induction was studied.The results showed that the best seed treatment to obtain aseptic seedlings was soaked in sterile water for 12 h,followed by disinfection with HgCl2 for 7min.Among the three explants,cotyledon was the most productive for the induction of callus,the premium medium was MS+0.8 mg/L 6-BA+0.10 mg/L IAA.
