摘要
应用分子生物学技术,选择鹦鹉热嗜衣原体(Chlamydophila psittaci,C.psittaci,Cps)6BC株的CPAF蛋白的免疫优势区基因,进行构建pGEX6p-2/CPAFm重组质粒与重组菌,使用IPTG诱导重组蛋白的表达并分析诱导温度、诱导剂剂量及诱导时间对蛋白表达的影响。重组蛋白以GST琼脂糖凝胶FF进行纯化,用SDS-PAGE和免疫印迹(Western blot)进行鉴定。为建立Cps的快速诊断和富集Cps诊断抗原奠定良好的试验基础。
It is selected the genes coding for immuno dominant region on CPAF protein from strain 6BC of Chlamydophila psittaci(Cps),which were used to construct pGEX6p-2/CPAFm recombinant plasmid and transformed E.coli BL21.The protein expression was induced by IPTG,the effects from temperature,dose and time of induction were investigated.The expressed protein was analyzed by SDS-PAGE and Western blot.The result showed that the recombinant antigen has a potential value for a sufficient antigen to be used in a rapid diagnosis for Cps infection.
