摘要 目的关节软骨损伤的早期诊断困难,血清中炎性因子、软骨基质金属蛋白酶类及Ⅱ型胶原代谢产物可作为关节软骨损伤的标志性产物。文中观察实验性兔关节软骨损伤早期血清中炎症因子白细胞介素-1(interleukin-1,IL-1)、基质金属蛋白酶-1(matrix metalloproteinase-1,MMP-1)、MMP-3、基质金属蛋白酶组织抑制因子-1(tissue inhibitor of matrix metallopro-teinase-1,TIMP-1)、Ⅱ型胶原降解产物Ⅱ型胶原羧基端端肽(collagen typeⅡC-telopeptide,CTX-Ⅱ)水平,探讨其在关节软骨损伤早期的分子生物学诊断意义。方法对新西兰白兔膝关节的股骨负重面实行人为创伤,用夹心ELISA法检测兔血清中IL-1、MMP-1、MMP-3、TIMP-1、CTX-Ⅱ水平。结果人为创伤性股骨软骨损伤的兔血清中IL-1、MMP-1、MMP-3、TIMP-1、CTX-Ⅱ水平均较对照组明显升高,差异有统计学意义。结论血清IL-1、MMP-1、MMP-3、TIMP-1、CTX-Ⅱ水平可作为早期关节软骨损伤的诊断指标。 Objective Articular cartilage injury is difficult to be diagnosed early.This study was to observe the levels of cytokine interleukin-1(IL-1),matrix metalloproteinase-1(MMP-1),matrix metalloproteinase-3(MMP-3),tissue inhibitor of matrix metalloproteinase-1(TIMP-1) and collagen type Ⅱ C-telopeptide(CTX-Ⅱ) in the serum of experimental rabbits with early articular cartilage injury,and investigate their molecular biological significance in the diagnosis of the early articular cartilage injury.MethodsMan-made trauma was inflicted on the knee femoral weight-bearing surface of New Zealand white rabbits,and ELISA was used to detect the levels of IL-1,MMP-1,MMP-3,TIMP-1 and CTX-Ⅱ in the serum of the rabbits.Results Statistically significant increases were found in the levels of serum IL-1,MMP-1,MMP-3,TIMP-1 and CTX-Ⅱ in the rabbits with man-made articular cartilage injury as compared with the controls.Conclusion The levels of serum IL-1,MMP-1,MMP-3,TIMP-1 and CTX-Ⅱ can serve as early diagnostic indicators of articular cartilage injury.
