火棘多糖清除氧自由基及抗脂质过氧化作用 被引量:12

Effects of Pyracantha Fortuneana Polysaccharide on Scavenging Oxygen Free Radical and Anti-lipid Peroxidation

摘要 目的:研究火棘多糖清除氧自由基与抗脂质过氧化作用。方法:利用邻苯三酚自氧化法产生超氧阴离子自由基(O2-.),采用Fenton体系产生羟自由基(OH.),研究火棘多糖对O2-.和OH.的清除作用;以Fe2+-H2O2系统诱导健康小鼠及四氯化碳(CCL4)肝损伤小鼠肝匀浆发生脂质过氧化,硫代巴比妥酸(TBA)法测定脂质过氧化产物丙二醛(MDA),研究火棘多糖的抗脂质过氧化作用。结果:火棘多糖可有效清除O2-.和OH.,可抑制Fe2+-H2O2诱导的健康小鼠肝匀浆脂质过氧化;火棘多糖不同剂量给药组能明显降低CCL4肝损伤小鼠肝脏内MDA的含量(P<0.01或P<0.05),火棘多糖给药组小鼠的肝匀浆能显著抑制体外诱导的脂质过氧化作用(P<0.01或P<0.05)。结论:火棘多糖具有清除氧自由基和抗脂质过氧化作用。 Objective To investigate the effects of Pyracantha fortuneana polysaccharide on scavenging oxygen free radical and anti-lipid peroxidation.Methods Superoxide anion free radical(O2-·) and hydroxyl free radical(OH·) were induced with auto-oxidation of pyrogallol and Fenton system,then the effects of Pyracantha fortuneana polysaccharide on scavenging O2-·and OH· were determined.The lipid peroxidation was induced with Fe2+-H2O2 system in liver homogenate of healthy rats and CCL4-induced acute hepatic injury rats,then the effects of Pyracantha fortuneana polysaccharide on anti-lipid peroxidation were determined,moreover,the production of lipid peroxidation malondialdehyde(MDA) was measured by thiobarabituric acid(TBA) colorimetric method.Results Pyracantha fortuneana polysaccharide could significantly scavenge O2-·and OH· and inhibit lipid peroxidation of healthy rat liver homogenate induced by Fe2+-H2O2 system.The different dosage of Pyracantha fortuneana polysaccharide could significantly inhibit the lipid peroxidation of liver homogenate from acute hepatic injury rats induced with CCL4 in vitro(P<0.01 and/or P<0.05),and decrease the contents of MDA in liver(P<0.01 and/or P<0.05).Conclusion Pyracantha fortuneana polysaccharide possesses the effects of scavenging oxygen free radical and anti-lipid peroxidation.
