摘要
目的:观察脂氧素受体激动剂BML-111对野百合碱(MCT)诱导肺动脉高压大鼠肺动脉压力的影响及其机制。方法:选择SD大鼠30只,随机分为3组:C组、PH组、B组,PH组和B组大鼠行皮下注射野百合碱50μg/g,C组注射等量0.9%氯化钠溶液,自野百合碱注射的当日起,B组每天腹腔注射1 mg.kg-1.d-1BML-111连续4周。实验测量指标:平均肺动脉压力(MPAP)、右心室收缩压(RVSP)、右心室(RV)[左室(LV)+室间隔(S)]、肺组织匀浆炎性因子白介素-8(IL-8)、内皮间粘附分子(ICAM-1)、单核细胞趋化蛋白(MCP-1)的水平。结果:PH组、B组MPAP、RVSP、RV/(LV+S)、IL-8、ICAM-1、MCP-1的水平高于C组(P<0.05);B组MPAP、RVSP、RV/(LV+S)、IL-8、ICAM-1、MCP-1低于PH组(P<0.05)。结论:脂氧素受体激动剂BML-111可减轻野百合碱诱导肺动脉高压大鼠肺动脉压力,其机制可能与减轻肺周围组织炎症反应有关。
Objective To observe the the effects of BML-111 on pulmonary arterial hypertension rats induced by monocrotaline and its possible mechanisms.Methods Thirty SD rats were randomly divided into group C,group PH and group B.The rats in group PH and group B were given subcutaneous injection with monocrotaline at a dose of 50 μg/g,while group C were given normal saline with the same volume.The rats in group B were given intraperitoneal injection with BML-111 at a dose of 1 mg·kg-1 daily for 4 weeks.The levels of MPAP,RVSP,RV/(LV+S),the concentrations of IL-8,ICAM-1,MCP-1 of pulmonary tissues were mearsured.Results The levels of MPAP,RVSP RV/(LV+S) and the concentrations of IL-8,ICAM-1,MCP-1 in group PH and group B were significantly higher than those of group C(P<0.05),meanwhile,the above indexes of group B were significantly higher than those of group PH(P<0.05).Conclusion BML-111 can ameliorate pulmonary arterial hypertension of rats induced by monocrotaline,wihch may relate to its reducing the inflammation of pulmonary tissues.
