摘要
利用黄酮类化合物可与金属离子形成有色螯合物的原理,建立柿叶中总黄酮含量测定方法,并且对供试品制备方法进行选择。通过正交试验设计选取最佳供试品制备条件,采用分光光度法,KAc-AlCl3体系与供试品中黄酮提取物反应,选择最大吸收波长,以芦丁为对照品制作标准曲线,并做方法学考察。供试品制备:提取液70%乙醇溶液,提取时间75 min,供试品与乙醇溶液的固液比为1∶50(g/mL),用KAc-AlCl3体系比色法测定供试品柿叶中总黄酮含量为1.13%,重复性RSD=1.86%(n=6),平均回收率为99.23%,RSD=0.27%(n=5)。KAc-AlCl3体系比色法测定柿叶中总黄酮含量的方法可行,呈色稳定,操作简便,可作为柿叶中总黄酮的含量测定方法,并可以做为快速测定方法应用于生产中对总黄酮含量的监控。
To establish the methods of determination of total flavones in diospyros leaves according to colorful chelate formed by flavone and metal ion and choose the optima conditions on sample preparation.Optima conditions of sample preparation were determined by mating design;spectrophotometry was conducted;KAc-AlCl3 system was reacted with flavones extract from samples;the maximum wavelength was selected;standard curve was made with Rutin-the control article;and methodology was investigated.Sample preparation was under the condition of 70 % ethanol,extraction 75 min,and the ratio of solid to liquid was;the content of total flavones was 1.13 % in diospyros leaves via KAc-AlCl3 system colorimetry and RSD was 1.86 %(n=6);the average recovery rate was 99.23 % and RSD was 0.27 %(n=5).The method of determination of total flavone in diospyros leaves via KAc-AlCl3 system colorimetry was feasible with stable color and simple conduction,which can be used for the determination of the total flavonoids and be suitable for quick quality monitoring of diospyros leaves.
