摘要
目的蝎氯毒素(Cltx)能特异性阻断神经胶质瘤细胞的氯离子通道,Cltx类似物(BmK CT)是从东亚钳蝎中纯化出来的第1个Cltx类似物。该文旨在研究BmK CT对大鼠胶质瘤细胞(C6)侵袭的影响及作用机制。方法采用Transwell小室评价BmK CT对C6细胞侵袭能力的影响,采用实时荧光定量聚合酶链反应(PCR)和酶联免疫吸附试验(ELISA)检测BmK CT对基质金属蛋白酶2(MMP2)基因和蛋白表达水平的影响。结果 BmK CT可显著抑制C6细胞的侵袭能力(P<0.05);C6细胞经BmK CT处理后,MMP2的mRNA表达未见明显变化,差异无统计学意义(P>0.05);但MMP2的分泌受到显著抑制(P<0.05)。结论 BmK CT可显著抑制C6细胞的侵袭能力,其机制可能与MMP2分泌水平降低有关。
Objective Malignant gliomas were the most common primary intracranial tumors in human.Chlorotoxin(Cltx) had been shown to selectively inhibit Cl-channels in glioma cells.Buthus martensi Karsch CT(BmK CT),the first chlorotoxin-like peptide cloned from the venom of BmK,had been a new research hotspot in the therapy of glioma.The present paper aimed to explore the effect and mechanism of BmK CT on the invasive ability of glioma cells.Methods Transwell method was used to evaluate the invasive ability of glioma cells.The real-time polymerase chain reaction(PCR) and enzyme-linked immunosorbent assay(ELISA) were used to detect the expressions of matrix metalloproteinase 2(MMP2) mRNA and protein.Result BmK CT significantly inhibited the invasive ability of glioma cells(P<0.05).The expression of MMP2 mRNA was not significantly different between groups with different BmK CT concentrations(P>0.05).The expression of MMP2 protein was inhibited by BmK CT significantly(P<0.05).Conclusion BmK CT can inhibit the invasive ability of glioma cells,which may be related with the decreased expression of MMP2 protein.
