摘要
采用正相硅胶柱层析和反相SG-64制备色谱柱,建立了从西洋参总皂苷中分离、制备人参皂苷类物质的方法。硅胶柱色谱分离采用三氯甲烷-甲醇为流动相梯度洗脱,反相SG-64制备色谱纯化采用甲醇-水梯度洗脱。分离过程采用薄层层析法和高效液相色谱法对目标化合物定性检测。分离得到6个纯度超过95%的单体人参皂苷类物质,根据单体化合物的理化性质和波谱数据对其进行结构鉴定,分别鉴定为人参皂苷Rg1、人参皂苷La、人参皂苷Rg2、人参皂苷Re、拟人参皂苷P-F11、人参皂苷Rd。结果表明,该方法操作简便,可用于西洋参皂苷化合物的分离纯化。
A method combining the normal phase silica gel column chromatography and reversed ph-ase SG-64 preparative liquid chromatography was developed to separate ginsenoside from crude saponins of Panax quinquefolium.The mobile phase for silica gel was chloroform-methanol system and for reversed phase SG-64 was methanol-water system.Six ginsenosides higher than 95% in purity were isolatedand and identified as ginsenosides Rg1,La,Rg2,Re,P-F11(pseudo-ginsenoside),Rd by physicochemical properties and spectral data.
