肥胖与非肥胖小鼠间充质干细胞成脂分化 被引量:1

摘要 分离培养瘦素受体基因突变型(db/db)和野生型(Wild Type,WT)小鼠原代间充质干细胞(Mesenchymal StemCells,MSCs);采用油红O染色和异丙醇萃取法分析不同分化时期细胞中脂质积累和成脂诱导率;免疫印迹检测PPARγ和脂联素蛋白水平。结果表明,db/db小鼠MSCs通过下调脂联素和PPARγ的表达抑制成脂分化过程;db/db小鼠和WT小鼠的脂肪细胞分化过程存在差异。 To isolate,culture and induce mesenchymal stem cells(MSCs) of the db deficient(db/db) and wild type(WT) mice,we use oil red O staining and isopropanol alcohol extraction method to analyze morphologic characteristic and lipid accumulation into the cells,and detect the expression levels of PPARγ and adiponectin by western blot.The results show that the MSCs of db/db mice could downregulate the expression of PPARγ and adipogenetic factors,and inhibit adipogenic differentiation of MSCs by some ways,and that the adipocyte differentiation process has some differences between obese mice and non-obese mice.
