氯化锂对脂肪组织Wnt信号及PPARγ表达的影响 被引量:1

摘要 对肥胖小鼠腹腔注射剂量为100和200 mg/kg小鼠体质量的氯化锂水溶液,实验期间记录小鼠体质量,于注射3周后采用半定量PCR和Western blot方法检测小鼠腹股沟处内脏脂肪组织中,β-catenin和PPARγ的表达变化。结果表明:氯化锂处理后小鼠体质量增长速率略有下降,β-catenin的mRNA水平无明显改变(p>0.05),蛋白水平明显上升(p<0.05),PPARγ的mRNA和蛋白水平表达均下降(p<0.05)。提示氯化锂可能通过上调β-catenin蛋白水平,并抑制PPARγ表达影响小鼠早期肥胖发生。 We investigate the effect of lithium chloride on the early stage of obesity.The experimental process is as follows: Inject either 100 mg/kg or 200 mg/kg of lithium chloride solution into each one of mice for three weeks;record the mouse body weight during the experimental period;check the expression of β-catenin and PPARγ by the methods semi-quantitative RT-PCR and Western blot;Use saline treatment group as a control group.Experimental results indicate that for the lithium-chloride-treated mice,the growth rate of body weight decreases slightly,the expression of β-catenin mRNA has no significant change(p>0.05) but its protein level rises significantly(p<0.05),and the expression of PPARγ decreases(p<0.05) both in mRNA and protein levels.These results suggest that lithium chloride may affect the early stage of obesity through increasing of β-catenin protein level and inhibiting of PPARγ expression.
