雌激素及精氨酸对去卵巢大鼠血清一氧化氮水平的影响 被引量:3

Effects of estrogen replacement and L-arginine supplementation on serum levels of nitric oxide in ovariectomized rats

摘要 目的 观察雌激素及精氨酸补充治疗对去卵巢大鼠血清一氧化氮(NO)水平的影响。方法 3月龄雌性Wistar大鼠40只,随机分为4组,A组:假手术组;B组:去卵巢组;C组:雌激素治疗组;D组:雌激素+L-精氨酸(L-arginine,L-Arg)治疗组。4月后用硝酸还原酶法测NO-3/NO-2的浓度来反映血清NO的浓度,并分别用放射免疫技术和高效液相色谱法检测血清中的雌二醇(E2)和L-Arg含量。结果 (1)和A组、C组、D组相比,B组E2和NO浓度显著降低,差异有统计学意义(P<0.05)。(2)C组和D组、A组相比E2浓度差异无统计学意义(P>0.05)。(3)C组和A组相比NO浓度无统计学差异(P>0.05)。(4)D组和C组相比NO浓度升高有统计学差异(P<0.05)。结论 E2可增加去卵巢大鼠血清NO水平。外源性L-Arg补充可提高血清NO水平。 Objective To investigate the effects of estrogen replacement and L-arginine(L-Arg) supplementation on serum levels of nitric oxide in ovariectomized rats.Methods Forty female Wistar rats,3-month-old,were randomly divided into four goups,goup A:sham-ovariectomized rats;group B:ovariectomized rats;group C:ovariectomized rats receiving estrogen replacement therapy;group D:ovariectomized rats receiving estrogen replacement and L-Arg supplementation therapy.Four months later,the concentrations of serum NO,E2 and L-Arg were assayed by nitrate reductase method,RIA and HPLC,respectively.Results(1) The concentrations of serum NO and E2 in group B was decreased significantly compared with other groups(P<0.05).(2) The serum levels of E2 had no significant difference in group A,C and D(P>0.05).(3) The serum levels of NO had no significant difference in group C and A(P>0.05).(4) The serum levels of NO in group D was increased significantly compared with group C(P<0.05).Conclusions(1) Estrogen could increase the concentration of serum NO in ovariectomized rats.(2) Exogenous L-Argsupplement could increase the concentration of serum NO in ovariectomized rats.
