天麻素治疗帕金森病大鼠的实验研究 被引量:14

摘要 目的探讨天麻素对治疗帕金森病(PD)大鼠神经行为及对中脑被盖腹侧区酪氨酸羟化酶(tyrosine hydroxylase,TH)阳性神经元的影响。方法采用6-羟多巴胺(6-OHDA)局部注射损毁右侧中脑腹侧被盖区(VTA),制备6-OHDA大鼠PD模型。40只造模PD大鼠分为对照组、左旋多巴组、天麻素组和左旋多巴与天麻素联合处理组(联合处理组),每组10只。观察PD大鼠旋转行为和用免疫组织化学法检测VTA区TH阳性神经元表达量。结果与对照组比较,3个治疗组PD大鼠旋转圈数均减少(P<0.05),PD大鼠VTA区TH表达量增加(P<0.05)。结论天麻素能明显改善PD大鼠旋转行为,增加PD大鼠VTA区TH表达,对TH阳性神经元有保护作用,天麻素和左旋多巴两药联用可能使这种作用增强。 Objective To explore the effect of gastrodin on neurobehavior and TH-positive neurons of ventral tegmentum area(VTA) in rat with Parkinson's disease(PD).Methods Rat models of PD were established by injecting 6-OHDA to the right midbrain tegmental ventral area.Forty rat models of PD were randomly divided into 4 groups:control group,L-dopar group,gastordin group and gastrodin combined with L-dapar group(n=10 in each group).The rotational behavior was observed and expressions of TH-positive neurons in ventral tegmentum area were detected by immunohistochemistry.Results Compared with control group,the number of rotation was reduced significantly(P<0.05) and amounts of TH-positive neurons were increased significantly(P<0.05) in three treatment groups.Conclusions Gastrodin can improve rotation behavior of PD rats,increase the expressions of TH-positive neurons in VTA and show a protective effect on TH-positive neurons.Gastrodin combined with L-dopar may enhance this effect much stronger.
