α-硫辛酸通过激活ERK1/2减轻葡萄糖/葡萄糖氧化酶所致心肌细胞损伤 被引量:1

摘要 目的探讨α-硫辛酸(LA)对葡萄糖/葡萄糖氧化酶(DG/GO)所致心肌细胞损伤的保护机制,着重探讨ERK1/2在其中的作用。方法将培养的大鼠H9c2心肌细胞分为如下几组:对照组、LA组、DG/GO组、LA+DG/GO组、PD+LA组、PD+LA+DG/GO组,3h后收集细胞测定ERK1/2磷酸化激活、24h测定细胞存活率。结果 LA显著提高ERK1/2磷酸化水平;PD98059完全阻断LA对ERK1/2的激活作用;以PD98059阻断LA对ERK1/2的激活后,完全去除LA对DG/GO所致细胞损伤的保护作用。结论 LA对DG/GO致心肌细胞损伤有保护性作用,是通过激活ERK1/2依赖性信号通路介导的。 Objective To investigate the role of ERK1/2 in the α-lipoic acid (LA)-induced cytoprotection against glucose/glucose oxidase (DG/GO) challenge in rat cardiac H9c2 myoblasts. Methods Rat H9c2 cells were randomly assigned into the following groups:control group, LA group, DG/GO group,LA+DG/GO group, PD+LA group, PD+LA+DG/GO group. ERK1/2 activation was analyzed 4 h after DG/GO challenge. Cell viability was determined 24 h after DG/GO stimulation. Results LA significantly increased the phosphorylation level of ERK1/2 compared with control group. PD98059 completely blocked LA-induced ERK1/2 activation. Blockade of ERK1/2 activation with PD98059 completely abolished the LA-induced increase in cell viability and decrease in apoptosis following DG/GO stimulation. Conclusions LA protects H9c2 cells from DG/GO-induced injury through ERK1/2 activation.
