摘要
目的研究福寿螺多糖对乙肝病毒转基因小鼠的作用。方法采用乙肝病毒转基因小鼠为模型,随机分为5组(每组8只):福寿螺多糖低、中、高3个剂量组,并设拉米呋啶组和对照组进行比较,均采用腹腔注射给药。观察用药前与用药后5d、10d、15d、20d及停药后5d小鼠HBV-DNA的表达。结果福寿螺多糖中、高剂量组在给药第15d、20d后,低剂量组在给药20d后,HBV-DNA拷贝量对比给药前均有显著性下降(1)P<0.05,2)P<0.01)。高剂量组在停药5d后HBV-DNA抑制率为15.76%与对照组相比有最著性提高。结论福寿螺多糖具有降低血清HBV-DNA含量,抑制乙肝病毒复制作用。
Objective To study the effect of AmpuUaria Gigas Spix on hepatits B virus genome transgenic mouse.Methods The transgenic mice of 6—10 weeks old were used as the animal models and randomly divided into five groups,low,middle,high dosage of AmpuUaria Gigas Spix group,lamivudine and control group.The drugs were intraperitoneal injected to the mice.The content of HBV—DNA was detected by PCR test at the pre-treatment,post-treatment and after withdrawal of treatment,respectively.Results The contents of HBV-DNA in the middle and high dosage groups after treated for 15 days,20 days,and in low dosage group treated for 20 days were obviously lower than that before treatment(1)P<0.05,2)P<0.01).The inhibition rate of HBV-DNA was 15.76%in high-dosage group in the 5th day after withdrawal of treatment,and increased remarkably compared with the control group.Conclusion AmpuUaria Gigas Spix polysaccharide can decrease the contents of HBV-DNA in serum and inhibit the replication of Hepatitis B virus.
