摘要
目的:采用HPLC-DAD-ELSD联用法对五种黄草石斛的指纹图谱进行对比研究。方法:采用DikmaC18(250mm×4.6mm,5μm)色谱柱;乙腈-0.5%冰乙酸水溶液梯度洗脱;柱温:30℃;流速:1mL.min-1;DAD串联蒸发光散射检测器检测。结果:建立了五种黄草石斛的HPLC指纹图谱,并标定了8个指纹峰。相似度结果表明,各种石斛相似度较低,差别较大。结论:建立的指纹图谱方法稳定、可靠、重现性好,可直接了解石斛品种与化学成分变化的关系。
Objective: To set up the fingerprint chromatograms of chemical principles of five kinds of Dendrobii Caulis by HPLC-DAD-ELSD. Method: Analysis was performed at 30℃ on a Dikma Cl8 column(4.6 mm×250 mm,5 μm) with a mobile phase consisting of acetonitrile-0.5% glacial acetic acid, gradient eluted at the flow rate of 1.0 mL·min-1 with DAD and ELSD detection. Result: HPLC fingerprint of Dendrobii Caulis was established, and 8 peaks were detected based on it. Similarity among different species was low. Conclusion: The method is stable, reliable and repeatable. It’s useful for the study on the relationship between chemical components and species of Dendrobii Cauli.
