黄芪多糖对实验性精索静脉曲张大鼠血CO浓度及睾丸细微结构影响 被引量:13

The influence of astragalus polysaccharide on CO concentration in blood and testicular micro-structure of varicocele rats

摘要 目的:研究黄芪多糖对实验性精索静脉曲张(Varicocele,VC)大鼠精索内静脉血中一氧化碳(CO)浓度及睾丸曲细精管结构影响,探讨黄芪有效成份治疗VC的作用及机制。方法:青春期雄性SD大鼠70只,随机分为假手术组(n=10,C组)和VC造模组(n=60)。其中,VC造模组大鼠按灌胃药物随机分为黄芪多糖实验组(n=40,A组)、生理盐水对照组(n=20,B组)。造模后分别于4周、8周取VC组和假手术组大鼠左侧睾丸,左侧精索内静脉血液。光镜结合图像分析系统观察睾丸组织形态变化;使用ELISA法检测血清中CO的吸光度。结果:各组间大鼠睾丸曲细精管内径、生精上皮高度比较均有统计学差异(P<0.05)。黄芪多糖治疗组损伤程度明显低于生理盐水组(P<0.01)。造模各组大鼠左侧精索内静脉血液中CO吸光度均有升高。黄芪多糖组CO吸光度低于生理盐水组,组间比较有统计学差异(P<0.01)。结论:实验性VC大鼠左侧精索内静脉血中存在CO浓度上调,其浓度变化可反映VC所致生精功能障碍程度,提示CO可作为临床早期判断VC患者生精组织损伤的预测性指标。黄芪多糖可改善由VC导致的睾丸生精组织的病理损伤,并且与用药时间呈正相关。黄芪多糖能降低VC所致大鼠血中CO的浓度。 Objective: To investigate the influence of astragalus polysaccharides(APS) on CO concentration in blood and testicular micro-structure of varicocele(VC) rats,and explore the mechanism of Astragali Radix in the treatment of varicocele.Method: Seventy adolescent male SD rats were randomly assigned to sham-operation group(n=10,C group) and varicocele(VC) model group(n=60).The 60 rats in model group were randomly assigned to astragalus polysaccharides group(n=40,A group),saline control group(n=20,B group).The left testis and blood from spermatic vein of VC group and sham-operation group were collected after 4 weeks and 8 weeks,respectively.The absorbance of CO was detected with ELISA method and the morphological changes of left testis were observed with microscope.Result: There was statistic difference in the injury of inside diameter of contorted seminiferous tubules,altitude of seminiferous epithelium among each group(P<0.05).The injury of seminiferous tubules in the APS groups was inferior to control group(P<0.01).The absorbance of CO in spermatic vein blood was up-regulated in the model rats.The absorbance of CO in APS groups was inferior to saline group.There was statistic difference of the absorbance of CO in each group(P<0.01).Conclusion: The concentration of CO is up-regulated in the blood from left spermatic vein of rats with varicocele.The concentration change can reflect the dysfunction caused by varicocele,indicating that CO can be used as the early clinical indicator in VC patients to predict the damage of the testis tissue.APS shows a time-relative effect in protecting body from testis damage caused by varicocele.Astragalus polysaccharides can reduce CO concentration in blood of VC rats.
