益智仁提取物对小鼠生精能力的影响实验研究 被引量:1

摘要 目的探讨益智仁提取物对环磷酰胺致生精障碍小鼠的治疗作用。方法水提法获得益智仁提取物。50只Balb/c小鼠随机分为对照组、模型组、益智仁提取物低剂量组、中剂量组和高剂量组(0.2g/kg、0.4g/kg、0.8g/kg)共5组。除对照组外,其余各组每天注射环磷酰胺75 mg/kg,连续5 d,制备生精障碍小鼠模型。末次给药后,益智仁提取物低剂量组、中剂量组、高剂量组分别给予相应剂量灌胃,对照组和模型组给予等体积生理盐水灌胃,每天1次,连续30 d。末次灌胃后24 h,取小鼠睾丸和附睾组织。检测各组小鼠精子密度、活率及精子畸形率,HE染色观察睾丸组织形态。结果与模型组相比,益智仁提取物各剂量组均可提高精子密度、精子活率,降低精子畸形率,组织学观察,益智仁提取物各剂量组小鼠睾丸组织病理性损害均得到改善。结论益智仁提取物对环磷酰胺所致小鼠生精障碍具有明显的治疗作用。 Objective To investigate the effect of sharpleaf galanal fruit extract on dyszoospermia induced by cyclophosphamide in mice.Methods Sharpleaf galanal fruit extract was extracted by ethanol-alkali solution.Fifty mice were randomly divided into 5 groups:model group,control group,low dose sharpleaf galanal fruit extract group(0.2 g/kg),middle dose sharpleaf galanal fruit extract group(0.4 g/kg),high dose sharpleaf galanal fruit extract group(0.8 g/kg).The dyszoospermia model was established by peritoneal injection of cyclophosphamide 75 mg/kg once a day for 5 days.Corresponding dose of sharpleaf galanal fruit extract was intragastric administered once a day for 30 days after last peritoneal injection.The control group and model group were intragastric administered equal-volume physiological saline.The testis were separated after 24 hours of last intragastric administration.Sperm count,motility rate and malformation rate were evaluated and testis morphology were determined.Results The sperm count and motility rate were higher and malformation rate was lower in each dose sharpleaf galanal fruit extract group than that in-model group.The pathological injury in testicular tissue was improved in each dose of sharpleaf galanal fruit extract group compared with the model group.Conclusion Sharpleaf galanal fruit extract has a therapeutical effect on dyszoospermia induced by cyclophosphamide in mice.
