摘要
目的研究hIL-10和HGF重组体对大鼠活体肝移植术后急性排斥反应的抑制作用。方法以DA大鼠(RT1a)为供体,Lewis大鼠(RT1l)为受体建立异种肝移植鼠模型,实验组于移植后1d及7d,腹腔内注射T-HGF和T-hIL-10重组体(100μg/mL)100μL,对照组注射等量生理盐水。移植后1、7、10和20d取大鼠尾静脉血,ELISA法测定血清HGF、IL-10水平,RT-PCR测定肝细胞HGF、IL-10mRNA的表达水平,常规肝功能测定,活体肝移植动物生存期观测。结果实验组鼠肝细胞有HGF和IL-10mRNA表达,对照组肝细胞表达微弱。对照组大鼠HGF和IL-10水平一直维持在较低水平,实验组大鼠HGF和IL-10水平明显高于对照组(P<0.05)。注射重组体的实验组平均生存时间为(19±0.9)d,20d时存活只数为6只,存活率75%;对照组平均生存时间为(8.6±0.5),20d时存活只数为2只,存活率25%(P<0.05)。对照组未注射重组体,ALT和AST值持续升高,注射重组体后ALT和AST值亦有升高,但明显低于对照组(P<0.05)。结论活体肝移植大鼠导入IL-10和HGF基因重组体后可抑制移植术后急性排斥反应。
【Objective】To study the suppression of hIL-10 and HGF on acute rejection in rats after orthotopic liver homeo transplantation (OLT). 【Methods】DA (RT1a) rats and Lewis (RT1l) were used as donors and receptors respectively. Treatment group was injected intraperitoneally with recombinant T-hIL-10 and T-HGF (100 μg/mL) 100 μL on day 1 and 7 after transplantation. Control group received the same dose of normal saline. The levels of HGF and IL-10 in serum were detected by ELISA,and HGF and IL-10 levels and their mRNA levels in hepato-cytes were detected by RT-PCR on day 1,7,10 and 20 after transplantation. Liver function was also detected by routine method,and the life span of liver transplantation rats was observed. 【Results】The levels of HGF and IL-10 in treatment group were higher than those in control group (P <0.05). There was HGF and IL-10 mRNA expression in hepatocytes of treatment group but not in those of control group. The mean survival time of treatment group was (19±0.9) days,and survival rats were 6 (75%) on day 20; while in control group,the mean survival time was (8.6± 0.5) days,and only 2 (25%) rats were alive on 20th day. In control group,the values of ALT and AST were increas-ing constantly. In treatment group,the values of ALT and AST were also increased,while the values were significant-ly lower than those of control group(P <0.05). 【Conclusion】The injection of recombinant IL-10 and HGF can sup-press the acute rejection after OLT.
