摘要
以高良姜的笋芽为试验材料,研究其表面消毒的方法及不同激素配比的培养基对高良姜笋芽的诱导、增殖和生根的影响。结果表明:用75%的酒精消毒10 s,无菌水漂洗1次,然后用0.1%的升汞浸泡10 min,无菌水漂洗3次的灭菌效果最佳,污染率可降至40%;适宜的芽增殖培养基为MS+8.0 mg/L 6-BA+0.5 mg/L NAA,增殖率达1.67;适宜的生根培养基为1/2MS+NAA 1 mg/L,生根率可达60%。
Taking Alpinia officinarum Hance's bud as experimental materials,this paper studies its surface sterilization and culture medium supplied with various hormone combinations have different effects on the induction,proliferation and rooting of alpinia officinarum hance. The result indicated: the most effective way of surface sterilization is achieved to disinfect with 75% alcohol for 10 s,rinse with sterile water for once,soak in 0. 1% mercuric chloride for 10 min,rinse with sterile water 3 times; suitable culture medium for bud propagation is MS+ 8. 0 mg / L 6-BA+ 0. 5 mg / L NAA,the proliferation rate achieves 1. 67; suitable culture medium for taking root is 1 /2MS+ NAA the 1 mg / L,rooting rate can achieve 60%.
