槲皮素-锌抗肿瘤作用及对HepG22的细胞凋亡诱导机制研究 被引量:8

摘要 [目的]观察槲皮素-锌配合物的体内外抗肿瘤作用及其机制。[方法]采用四氮唑盐(MTT)法观察槲皮素-锌配合物对人肝癌细胞HepG2的抑制率,并采用流式细胞术(FCM)观察分析癌细胞周期;通过建立荷瘤小鼠模型,观察槲皮素-锌配合物对S180荷瘤小鼠瘤重和H22小鼠生存时间的影响,观察其体内抗肿瘤作用。[结果]槲皮素-锌配合物可显著抑制HepG2的体外增殖,且抑制作用呈浓度依赖性,可使G0/G1期细胞增多,S期和G2/M期细胞减少。对S180小鼠的瘤体生长有显著的抑制作用,对H22小鼠的生存时间有显著的延长作用。[结论]槲皮素-锌配合物在体内、外均能有效抑制肿瘤细胞的增殖,并诱导HepG2肿瘤细胞的凋亡。 [Objective]To study the antitumor functions and tumor cell apoptosis induced by Quercetin Zn.[Methods]MTT assay was applied to study the antitumor activities of quercetin Zn in HepG2 cell lines in vitro,and apoptotic cell was analyzed by flow cytometry.The in vivo antitumor action was studied by observing the variation of tumor weight and survival time of S180 and H22 mice.[Results]Quercetin Zn can inhibit HepG2 cells from growth and proliferation,and evoke apoptosis.Flow cytometry showed that it caused an increase at G0/G1 phase and a decrease at G2/M phase and arrest at G0/G1 phase in the cell cycle.It also can inhibit the growth of cancer cells of S180 mice and prolong the survival time of H22mice.[Conclusion]Quercetin Zn has antiproliferative and proapoptotic effect on tumor cells in vivo and in vitro.
