摘要
收集妊娠 2 6 .0~ 2 9.0d的猪胚胎 ,分离培养原始生殖细胞 (PGCs) ,在STO细胞饲养层上生长 ,形成了多能性干细胞 (EG)。发现其具有干细胞的显著特性 ,并能在体外定向分化为神经细胞、上皮细胞和胚泡细胞。试验使用A、B、C 3种不同的培养基培养PGCs ,结果在形成EG细胞集落的数量和EG细胞分化上有着明显的差距 。
Embryonic germ(EG) cells are undifferentiated stem cells isolated from cultured primordial germ cells( PGCs). PGCs collected from day 26 or 29 porcine embryos were cultured on mitotically inactivated murine fibroblasts(STO). EG cells share several characteristics with embryonic stem(ES) cells, (including) morphology, pluripotency and the capacity for germline transmission. EG cells possessed alkaline phosphatase activity,and differentiated into several cell types in vitro. PGCs were cultured respectively with the different culture medium. These results suggested that the quality and concentration of serum ,and addition of growth factors are essential for the in vitro survival and proliferation of cultured porcine PGC.
