本文以[~3H]地塞米松(Dex)为配体,用放射配体结合测定法研究了80天游泳应激(1次/天,40~50分/次)后大鼠肝、脑胞液和胸腺细胞糖皮质激素受体(GR)的变化,与对照组比较,游泳组大鼠肝胞液,胸腺细胞的[~3H]Dex特异结合量减少,表现上的平衡解离常数Kd不变,脑胞液GR变化不明显。实验结果提示,慢性应激可导致GR减少。对这些变化的可能的机制和生理意义作了讨论。
Using ~3H-dexamethasone as ligand, changes of glucocorticoid receptors (GR) in rat liver and brain cytosol and thymocytes were studied with radioligand binding assay. The rats underwent swimming training 40-50 min/day for 80 days. Results indicate that the binding capacity (Ro) of GR in hepatic cytosol and the specific binding (Rs) of GR in thymocytes were significantly lower in exercise group than in the controls, but the apparant dissociation constant (Kd) was not changed. The GR in brain cytosol showed no significant reduction after training. These findings show that chronic stress can result in a decrease in GR. The possible mechanism and physiological significance of these changes were discussed.
