摘要 目的:研究不同提取温度、提取方法对紫草中有效成分的影响,优选紫草中萘醌类成分的提取工艺。方法:采用分光光度法测定紫草提取液中总萘醌含量,比较渗漉、回流、温浸(65℃)、超声、浸渍方法的提取效率,采用L9(34)正交试验,以总萘醌的含量作为指标,以乙醇用量、浸泡时间和浸泡次数为考察因素,优选紫草的醇提工艺条件。结果:回流对萘醌类成分的破坏明显,其余4种提取方法以渗漉法、温浸法(65℃)提取率最高,各方法之间无显著差别;优选的醇提工艺为:加10倍量95%乙醇,浸泡2次,每次12小时。结论:浸渍法节约能源,对设备要求低,操作简便,转移率高,优选的提取工艺稳定、合理、可行,适用于工业化生产。 Objective: Study on the influence of the different extraction temperature and extraction method for effective components in Radix Arnebiae to select the optimal extraction process of naphthoquinone in Radix Arnebiae. Method: Spectrophotometry was used to detect the naphthoquinone in Radix Arnebiae and the extract rates by percolation, reflux, soak(65℃), ultrasonic and dip method were compared.With L9(3)4 orthogonal test, the alcohol extraction conditions of Radix Arnebiae was optimized with naphthoquinone content as the index, and the amount of alcohol, extraction time, extraction times as the investigation factors. Result: Among the five methods, naphthoquinone was destroyed by percolation obviously. The extraction rate of naphthoquinone were higher in sample prepared by soak(65℃) and dip methods with no significant difference among the preparation methods. The optimal alcohol extraction conditions were as following:dipping extracting 2 times with 10-fold 95% alcohol, 12 hours for each time. Conclusion: The dip method is simple and save energy and has lower demands for equipments. The optimum extraction process is stable, feasible and reasonable.
