摘要
本研究利用龙眼(Dimocarpus longana Lour)大叶系的“东壁”及小叶系的“红核子”的幼子叶,诱导出大量肾形胚。这些肾形胚发育成正常试管苗,试管苗已经土培成活。在愈伤组织诱导中,以MS为基本培养基,补加K 1.0 mg/L,2,4-D 2.0 mg/L及0.5%活性炭效果最好,进而可在附加0.5 mg/L 6 BA和0.1mg/L NAA下分化出肾形胚,在较低水平激素及附加必要有机物情况下,肾形胚可以大量繁殖继代。并可继续萌发成正常幼苗。从我们的实验结果看,有可能利用诱导的胚状体继代作为无性系,以便为性状正齐一致的,优良的龙眼砧木的快速繁殖开创新途径,并可作为育种的一种手段。
In present study,agreat number of reniform embryoids were induced from the cotyledons of two strains of Dimocarpus longana Lour.,the large-leaf strain 'Dong-pi' and the small-leaf strain 'Hung-he-zi'. These embryoids have been grown into normal test-tube plant,which have survived in the soil. The MS medium with additional kinetin 1.0mg/L,2-2-4-D 2.0mg/L,and 0.5% activated carbon showed a satisfactory result for the induction of callus from cotyledon. Calli differentiated into reniform embryoids when BA 0.5 mg/L and NAA 0.1mg/L were added to the medium. Reniform embryoids could propagate themself in large quantities and could develop into normal plants when a lower level of hormone and additional essential organic matter were present in the medium. The results show that it is possible to use the generation of induced embryoids as a clone for the rapid propagation of longan stock with good characters and for the selection of desirable type in longan breeding.
