HPLC法分析乳清蛋白对糖尿病模型小鼠血浆氨基酸组分的影响 被引量:1

摘要 为寻求有效的氨基酸分离方法并探讨乳清蛋白对2型糖尿病防治的作用机制,采用HPLC法分析乳清蛋白中氨基酸组分及含量;分别以0%、10%、20%和40%的乳清蛋白(WP)灌胃1型、2型糖尿病模型组、正常组小鼠,4周后观察各组血浆氨基酸的变化。乳清蛋白中亮氨酸、异亮氨酸、缬氨酸分别占氨基酸总量的14.40%、5.93%和5.32%,即支链氨基酸(BCAA)的含量为25.65%。无论是正常小鼠还是造模小鼠,乳清蛋白灌胃后血浆中支链氨基酸含量上升,并且灌胃浓度越高,BCAA含量越高。另外,造模小鼠血液中BCAA含量高于正常小鼠。乳清蛋白富含的BCAA灌胃后增加了模型小鼠血浆氨基酸水平。摄取乳清蛋白可增加小鼠血浆中氨基酸水平尤其是BCAA水平。 To seek the effective method of extraction of amino acids and discuss about the mechanism of whey protein on prevention and control of type 2diabetes,HPLC method was used to analyze the amino acid composition and content in the whey protein.Assigned the mice at random in four groups and administered intragastrically with 0%,10%,20%and 40%of the whey protein for 4weeks,according to the HPLC measurement,leucine,isoleucine,and valine of whey protein were accounted for 14.40%,5.93%and 5.32% of the total amino acids respectively.It also meant branched-chain amino acids(BCAA)were accounted for 25.65%.After using the intragastrical way with whey protein,BCAA of plasma in both normal mice and model mice increased and the higher the concentration of gastric perfusion,the higher content the BCAA contained.In addition,content of BCAA in blood of model mice was higher than the normal mice and BCAA of whey protein increased the plasma amino acid level in model mice after administration.
