摘要
目的将H9N2亚型猪流感病毒(H9N2-SIV)进行增殖并测定其毒力。方法采用鸡胚尿囊腔增殖法,将病毒接种于10日龄SPF鸡胚进行增殖,血凝(HA)试验测定病毒液效价,Reed-Muench法测定病毒的鸡胚半数感染量(EID50)和小鼠半数致死量(MLD50)。结果经过增殖后,H9N2-SIV血凝效价为1∶256,EID50=10-6.5·0.1mL-1,MLD50=10-2.32·0.l mL-1。结论 H9N2-SIV可以作为进一步试验用病毒。
Objective To proliferate swine influenza virus (H9N2) (H9N2-SIV)and determine its virulence.Methods Using chicken embryo allantoic cavity inoculation,the 10 d SPF chicken embryo was inoculated to proliferate the virus,and titer of H9N2-SIV was determined by hemagglutination (HA)ex-periment.Reed-Muench method was used to determine EID50 and MLD50 of H9N2-SIV.Results Hemag-glutination titer of H9N2-SIV was 1∶256,EID50=10-6.5·0.1 mL-1,MLD50=10-2.32·0.l mL-1 after pro-liferation.Conclusion H9N2-SIV could serve as a further test virus.
