响应面法优化超临界CO_2法萃取废弃烟叶中类胡萝卜素的工艺条件 被引量:3

Optimization of Supercritical CO_2 Extraction Technology of Carotenoids in Waste Tobacco Leaf by Response Surface Methodology

摘要 采用超临界CO2法萃取废弃烟叶中类胡萝卜素,以类胡萝卜素提取率为指标,在单因素试验的基础上,采用Box-Behnken响应面法对萃取工艺的关键因素萃取压力、萃取温度、萃取时间和CO2流速进行优化。结果表明,萃取温度、萃取压力和CO2流速的交互作用对烟叶中类胡萝卜素的提取率影响极显著;萃取压力的影响显著。优化得到超临界CO2法提取烟叶中类胡萝卜素的最优条件为萃取压力23.53MPa、萃取时间1.72 h、萃取温度50.00℃、CO2流速8.05 L/h,此条件下烟叶中类胡萝卜素的提取率为285.1μg/100 g。 Carotenoids were extracted from waste tobacco leaf by supercritical CO2 extraction method,using the yield of carotenoids as indicator,the key factors of supercritical CO2 extraction method,extraction pressure,temperature,time and CO2 flow speed were optimized by Box-Behnken response surface methodology on the base of single factor tests.Results indicated that extraction temperature,the interaction between extraction pressure and CO2 flow speed had very significant effects on the yield of carotenoids;while extraction pressure had significant effects.The optimal extraction conditions for the extraction of tobacco carotenoid were obtained as followed,extraction pressure,23.53 MPa;extraction time,1.72 h;extraction temperature,50.00 ℃;CO2 flow speed,8.05 L/h,the yield of carotenoid under these conditions was 285.1 μg/100 g.
