摘要
为了探讨甲醛对小鼠造血功能的影响,将昆明小鼠30只随机分为5组,分别为低、中、高浓度甲醛组,盐酸苯肼试验组和空白对照组,试验期间连续腹腔用药,第二天起每天检测红细胞数量、白细胞数量、血红蛋白含量以及网织红细胞比例等参数的改变,试验第十天检测后处死小鼠取肝脏和骨髓常规石蜡切片HE染色。结果表明,用药第二天小鼠即出现溶血现象,第八天时,与空白对照组相比高浓度组小鼠红细胞数量降低了32.47%,血红蛋白含量降低了44.14%,网织红细胞比例高达22.86%;肝脏、骨髓切片可见较多破坏的红细胞。甲醛对小鼠有致溶血作用,能严重影响小鼠的造血功能。
30 Kunming mice were randomly divided into 5 groups such as the low, medium and high concentration groups of formaldehyde, phenylhydrazine hydrochloride group and negative control group to detect the effect of formaldehyde on mouse hematopoiesis. During the experiment, the experimental animal was continuously treated with formaldehyde or phenylhydrazine hydrochloride by daily intraperitoneal injection, and normal saline solution to negative control. Since the next day after injection, the quantity of red blood cell(RBC) and leukocyte, the content of hemoglobin and reticulocyte proportion were recorded daily. At the tenth day after injection, the mice were killed to sample liver and bone marrow after detection. The bone marrow were performed traditionally paraffin slice and H-E staining. The results showed that at the second day after injection(DAI), the mice injected with drugs showed the phenomenon of anemia, and at about eight DAI, the quantity of RBC and hemoglobin content of each group treated by drugs decreased by 32.47% and 44.14%, respectively, and the reticulocyte proportion was up to 22.86%. When treated with drugs sequentially, each index would maintain the same levels as above. Some broken RBC were viewed in the liver and bone marrow slice. It indicated that the formaldehyde could cause haemolysis in mouse, and seriously affect the mouse hematopoiesis.
