摘要 为了找出八仙花不同部位总RNA最佳的提取方法,分析花色素苷对RNA提取的影响,以八仙花的根、茎、叶、花,以及蓝白环、粉色环、红色花、蓝色花、紫色花及白色花6种不同颜色八仙花的花萼为材料,分别采用试剂盒法和Trizol法提取总RNA。试验结果表明:试剂盒法可获得较高质量的八仙花植物总RNA,OD260/OD280在1.9~2.1之间;在4个不同部位中,以八仙花叶片和花中RNA含量较高,且较易提取,在不同色的花萼中,以红色花花萼中RNA含量最高,且最易提取;用试剂盒法提取时,花色素苷对八仙花总RNA的提取并无显著影响。 In order to find the best extraction method for total RNA from different parts of Hydrangea macrophylla and analyze the influence of anthocyanin on total RNA extraction,the root,stem,leaf,flower of Hydrangea macrophylla and the calyxes from six different colors of Hydrangea macrophylla(blue-white ring,pink ring,red flower,blue flower,purple flower and white flower) were used as material to extract total RNA by kit method and Trizol method,respectively.The results showed that a better quality of total RNA can be extracted from Hydrangea macrophylla by the kit method,and the OD260 /OD280 was between 1.9~2.1;among the four different parts,the RNA content was high in leaf and flower with easier extraction;among different colors of calyx,the RNA content was the highest in calyx from red flower with the easiest extraction;with the kit method,anthocyanin had no significant influence on extraction of total RNA from Hydrangea macrophylla.
