失答刺知丸对脑缺血再灌注损伤大鼠血清hs-CRP及IL-6水平的影响 被引量:7

Effects of Shidalazhiwan on Serum hs-CRP and IL-6 in Rats with Cerebral Ischemia-reperfusion Injury

摘要 目的观察回族医药经典方药"失答剌知丸"对脑缺血再灌注大鼠模型血清超敏C-反应蛋白(hs-CRP)、白介素-6(IL-6)水平的影响。方法设假手术组、模型组、失答剌知丸组、盐酸氟桂利嗪胶囊组,后三组再根据不同时间点分四个亚组,即6h组、2d组、7d组、14d组,将156只健康雄性SD大鼠随机分13组,每组12只。造模方法参照改良的Zea longa法线栓阻塞大鼠大脑中动脉制备局灶性脑缺血再灌注动物模型,随后行神经行为功能测试以观察模型的可靠性,运用放射免疫法和免疫比浊法分别检测并比较血清中hs-CRP和IL-6的含量水平变化。结果①大鼠脑缺血再灌注损伤后有明显的神经行为功能缺失,模型组各时间点评分较假手术组均明显升高(P<0.01);与模型组比较,各药物组评分均显著下降(P<0.01);与氟桂利嗪组比较,失答剌知丸组2、7、14d组评分均显著下降(P<0.01)。除假手术组外,各组不同时间点间比较神经功能评分逐渐下降,失答剌知丸组较为明显(均P<0.05)。②大鼠脑缺血再灌注损伤后血清hs-CRP表达:与假手术组比较模型组均明显升高(P<0.01);与模型组比较,药物组氟桂利嗪7、14d、失答剌知丸组2、7、14d均显著下降(P<0.01);与氟桂利嗪组比较,失答剌知丸组2d组显著下降(P<0.01)。③大鼠脑缺血再灌注损伤后血清IL-6表达:与假手术组比较模型组均明显升高(P<0.01);与模型组比较,药物氟桂利嗪组7、14d均显著下降(P<0.01),失答剌知丸组2d明显下降(P<0.05),7、14d均显著下降(P<0.01)。结论失答剌知丸干预可促进脑缺血再灌注损伤大鼠神经行为功能缺损的恢复,其作用与降低血清炎性反应因子hs-CRP和IL-6水平有关。 Objective To observe the effects of SDLZW in classic medicine prescriptions on high - sensitivity c -reactive protein(hs -CRP) and IL -6 in cerebral ischemia - reperfusion injure rats model.Methods Rats were divided randomly into Sham,Model,SDLZW and Flunarizine Hydrochloride Capsules(FHC ).The later three groups were divided into four subgroups according to different points in time,namely 6h,2d,7d and 14d group,12 rats in each group(subgroup).Rat focal cerebral ischemia model was established by middle cerebral artery occlusion(MCAO) with nylon thread.Changes of serum hs - CRP and IL -6 were detected by the method of Immunoturbidimery and Radioimmunoassay.Results①After cerebral ischemia - reperfusion injury,rats showed significant neurobehavioral function missing,and scoring in model groups all time were significantly elevated than that in pseudo surgery group(P &lt;0.01).Compared with model group,FHC group and the SDLZW group showed significantly falling in scoring(P &lt;0.01).Except pseudo surgery group, the neurological function score decreased significantly with time going in each group.②The serum hs - CRP expression in rats:Compared with the model group,FHC 7d,14d group and the SDLZW 2d,7d,14d group showed falling significantly(P &lt;0.01).Compared with FHC group,SDLZW 2d group decreased significantly (P &lt;0.05).③The IL -6 expression in rat serum;Compared with the model group,FHC 7d,14d group and the SDLZW 2d,7d,14d group decreased significantly(P &lt;0.05).Conclusion SDLZW has the therapeutic effect on cerebral ischemia / reperfusion injury,which can reduce the serum expression of inflammatory factors hs - CRP and IL - 6 and also have a good recovery effect on neurobehavioral function in rats with defect.
