丙泊酚对大鼠急性脊髓损伤早期的影响 被引量:1

摘要 目的观察丙泊酚对大鼠急性脊髓损伤早期的影响,探讨丙泊酚对急性脊髓损伤早期脊髓神经组织的保护作用。方法健康雄性SD大鼠48只随机分为6组,每组8只。假手术组(S组)、脊髓损伤组(I组)、脂肪乳组(F组)和不同剂量丙泊酚组:低剂量组(L组)、中剂量组(M组)和高剂量组(H组)。S组仅切除T9全椎板,不损伤脊髓;其余各组应用Allen,s重物坠落法并加以改良制备大鼠急性脊髓损伤模型。各组药物干预15min(T15)和60min(T60)时取动脉血大鼠1.5mL测血清S100β蛋白含量,各组药物干预60min后立即断头处死大鼠取脊髓组织测脊髓含水量及脊髓组织总钙含量。结果与S组比较,其余五组在T15和T60时血清S100β蛋白和脊髓组织总钙含量均明显升高(P均<0.01),I组、F组、L组和H组脊髓含水量均增多(P均<0.05);I组与F组间各时间点血清S100β蛋白含量、脊髓组织含水量及总钙含量差异均无统计学意义(P>0.05);与I组和F组分别比较,在T15和T60时L组、M组和H组血清S100β蛋白和脊髓组织总钙含量均显著降低(P均<0.01),M组脊髓含水量低于I组和F组(P<0.05);与L组比较,在T15和T60时M组和H组血清S100β蛋白和脊髓组织总钙含量均降低(P均<0.01);与M组比较,在T15和T60时H组血清S100β蛋白和脊髓组织总钙含量均升高(P均<0.01),脊髓含水量增多(P<0.05);与T15时比较,L组、M组和H组在T60时血清S100β蛋白含量均明显降低(P均<0.01)。结论丙泊酚34.92mg·(kg·h)-1持续泵注60min对急性脊髓损伤后早期有保护作用。 Objective To investigate he effects of propofol on acute early spinal cord injury,and to explore the protective effect of propofol on the early acute spinal cord injury spinal cord nerve tissue. Methods 48 healthy male SD rats were randomly divided into six groups with 8 rats each( n = 8),sham group( group S), spinal cord injury group( group I),fat emulsion group( group F) and different concentrations of propofol groups( group L,M,H). T 9 laminectomy was resected with no damage to the spinal cord in group S. Acute spinal cord injury model was established with Allen’s fall method in rats of rest groups. At the drug intervention 15min( T 15) and 60min( T 60),1. 5mL blood samples were collected for determination S100β protein contents. The rats were sacrificed immediately after the drug intervention 60min. Spinal cord water contents and total calcium of spinal cord tissues were measured. Results Compared with group S,S100βprotein contents and total calcium of spinal cord tissue in the other five groups significantly increased at T 15 and T 60( P &lt;0. 01) while the spinal cord water content increased in groups I,S,L and H( P &lt; 0. 05); Compared with group I, S100βProtein content,total calcium of spinal cord tissue and the spinal cord water content no significant change were found in group F( P &gt; 0. 05); Compared with group I and F,S100βProtein content and total calcium of spinal cord tissue in groups L,M and H significantly decreased at T 15 and T 60( P &lt;0. 01) while the spinal cord water content decreased in group M( P &lt; 0. 05); Compared with group L,S100βProtein content and total calcium of spinal cord tissue in groups M and H decreased at T 15 and T 60( P &lt;0. 01); Compared with group M,S100βProtein content and total calcium of spinal cord tissue in group H increased at T 15 and T 60( P &lt;0. 01) while the spinal cord water content increased in group H( P &lt; 0. 05); Compared with T 15, S100βProtein content in groups L,M and H decreased at T60( P &lt; 0. 01). Conclusion Propofol[34. 92 mg·( kg·h)- 1 ]continuous infusion 60min has a protective effect on early acute spinal cord injury in rats.
