摘要 目的探讨建立实验性感音神经性耳聋动物模型的有效方法。方法40只小鼠平均分组,皮下注射新霉素及噪声暴露,8只小鼠作为对照组不予任何处理,致聋前及致聋后14d应用脑干诱发电位仪(ICS CHARTREP)在隔声屏蔽室内行双侧ABR阈值测试。各组小鼠于在进行ABR检测后,通过耳蜗基底膜铺片来检测毛细胞的病理损伤过程。结果与对照组比较,两组致聋组ABR阈值均显著增高,致聋后两组基底膜染色铺片比较,毛细胞损伤程度相近。结论两种方法均能造成SD大鼠感音神经性耳聋,但新霉素大剂量皮下注射与噪声刺激相比较操作更简便有效。 Objective To explore the effective method of establishing experimental animal models of sensorineural hearing loss.Methods 48mice were randomly divided into 3 groups.20 mice were exposed to 2-4KHz narrow band noise at 100dB SPL 6h per day for 3 consecutive days.20 mice were using subcutaneous injections of neomycin.The rest 8 mice served as the control group without receiving anything.To assess hearing threshold shift,auditory brainstem response(ABR) were examined at three group prior and after deafness.To detect the pathological damage process of hair cell with the surface preparation techniques of the cochlear basilar membrane after ABR detection.Results Compared with the control group,ABR threshold of the two experimental groups were significantly higher,and the damage degree of hair cells was similar between the two experimental groups after the surface preparation techniques of the cochlear basilar membrane.Conclusion Two methods both can cause the SD rat sensorineural hearing loss,but large dose subcutaneous injection of neomycin wiube simpler and more effective than narrow band noise exposure.
