响应曲面法优化发酵辅助提取葛根素的工艺研究

摘要 以葛根为原料,利用响应曲面法,以二次回归方程预测模型为基础,对黑曲霉发酵葛根的预处理条件进行了优化,将生物预处理与醇提工艺相结合,有效提高了葛根素的提取率。实验表明,曲面回归方程与实验结果拟合性好,优化预处理条件为:料液比1∶10,接种量10%,发酵温度29℃,发酵时间97h,醇提后葛根素提取率达到7.03%,是未经预处理的3.48倍;用红外光谱分析和薄层层析法对提取的葛根素进行表征,确定提取的产物为葛根素;对葛根素提取液的抑菌活性进行检测,结果表明葛根素的抑菌效果为:大肠杆菌>金黄色葡萄球菌>绿脓杆菌>白色念珠菌。 Using pueraria as raw material and combining biological pretreatment with alcohol extraction process, the extraction rate of puerarin was improved effectively.The response surface methodology(RSM)was adopted and the quadratic regression equation prediction model was used to optimize the pretreatment conditions of fermentation pueraria by Aspergillus niger.Experimental results showed that the fit of surface regression equation with the experimental results was good,optimized pretreatment conditions were as follows:the ratio of material to liquid of 1∶10, inoculum of 10%,fermentation temperature of 29℃,and fermentation time of 97h.After alcohol extraction,the extraction rate of puerarin was up to 7.03%and 3.48times of the control.The bacteriostatic experiments showed that the bacteriostatic effect of puerarin was:Escherichia coli>Staphylococcus aureus>Pseudomonas aeruginosa>Candida albicans.
