摘要
目的探讨2型糖尿病大鼠骨折愈合过程与体内血清中BMP-2、TNF-a、IL-1变化关系的研究。方法取清洁近交SD雄性大鼠随机分为两组;正常对照组(NC)和糖尿病组(DM),DM组以高脂高糖饲料喂养10周联合腹腔注射小剂量链脲佐菌素的方法建立2型糖尿病大鼠模型后,折断两组大鼠右胫骨并复位外固定。分别于骨折后第1、3、5、8周拍X线片观察骨折愈合情况;同时尾静脉取血并留取血清,用ELISA方法检测2组血清中BMP-2、TNF-a、IL-1水平。结果骨折后第1、3、5、8周X线片观察NC组骨折愈合好于DM组;DM组骨折后血清BMP-2浓度显著低于NC组,而血清TNF-a与IL-1浓度显著高于NC组。结论糖尿病大鼠骨折后血清BMP-2减少、TNF-a和IL-1增加与其骨折愈合差密切相关。
Objective To observe the concentration changes of serum BMP- 2,TNF- a,IL- 1 during fracture healing in rats with type 2 diabetes mellitus in vivo. Methods The clean inbred Sprague- daw Ley( SD) male rats were randomly divided into two groups: the normal control group( NC) and diabetic group( DM). The DM group rats were fed with high fat and sucrose diet for 10 weeks,then injected with small dose streptozotocin by intraperitoneal to establish the rat model of type 2 diabetes. The right tibia of two group's rats were broken,reset and externally fixed. The fracture healing was observed with X- ray,while serum levels of BMP- 2,TNF- a and IL- 1 were detected by ELISA. Results X- ray films showed that the fracture healing of normal control group was significantly better than that of diabetic group. In DM group,the levels of BMP- 2 decreased significantly( P < 0. 05 or P < 0. 01) as compared with NC group,while levels of TNF- a and IL- 1 increased significantly( P < 0. 05 or P < 0. 01) as compared with NC group. Conclusion The unfavorable fracture healing of rats with type 2 diabetes mellitus was associated with the BMP- 2 reduction and high serum levels of TNF- a and IL- 1.
