长链非编码RNA PVT1对MDA-MB-231细胞增殖和侵袭能力的影响 被引量:1

摘要 目的探讨长链非编码RNA PVT1(lncRNA-PVT1)对于乳腺癌细胞系MDA-MB-231增殖和侵袭的影响。方法构建pcDNA3.1-PVT1表达载体,并将其转染入MDA-MB-231细胞,用实时定量PCR方法检测转染细胞中lncRNA-PVT1的表达,用Edu标记技术检测转染细胞的增殖能力,用Transwell实验检测转染细胞的侵袭能力。结果构建成功的pcDNA3.1-PVT1重组质粒转染入MDAMB-231细胞后lncRNA-PVT1表达增加;转染细胞的增殖能力增强;转染细胞的侵袭能力增强。结论 lncRNA-PVT1能增强MDA-MB-231细胞的增殖和侵袭能力。 Objectives To study the effect of ectopic expression of long Noncoding RNA PVT1 in MDA-MB-231 cells. Methods The pcDNA3.1-PVT1 plasmids were transfected into MDA-MB-231 cells and the expression of lncRNA-PVT1 was detected by Real time PCR. The proliferation ability of transfected cells was evaluated by Edu based assay. The invasion ability of transfected cells was evaluated by Transwell experiment. Results The expression of lncRNA-PVT1 was increased when the recombinant plasmids were transfected into MDA-MB-231 cells. The proliferation ability of transfected cells was improved and so was the invasion ability. Conclusion Long noncoding RNA PVT1 can improve the proliferation and invasion of MDA-MB-231 cells.
