摘要
目的 探讨人脑胶质瘤血管内皮细胞生长因子 (VEGF)蛋白和mRNA的表达与微血管密度(MVD)的关系及其临床意义。 方法 4 5例人脑胶质瘤标本 ,其中星形细胞瘤 (Ⅰ级 )和星形母细胞瘤 (Ⅱ级 )标本 2 5例 ,间变型 (Ⅲ级 )和多形胶母细胞瘤 (Ⅳ级 )标本 2 0例 ,使用LsAB免疫组化方法检测胶质瘤标本的VEGF、内皮细胞相关抗原 (Ⅷ因子 )。使用原位杂交的方法检测VEGFmRNA的表达。 结果 Ⅰ级和Ⅱ级胶质瘤标本VEGF强阳性表达 3例 ,弱阳性表达 2 2例 ,Ⅲ级和Ⅳ级胶质瘤标本VEGF强阳性表达 18例 ,弱阳性表达 2例。Ⅰ级和Ⅱ级胶质瘤标本VEGFmRNA强阳性表达 2例 ,弱阳性表达 2 3例 ,Ⅲ级和Ⅳ级胶质瘤标本VEGFmRNA强阳性表达 17例 ,弱阳性表达 3例。Ⅰ级和Ⅱ级胶质瘤标本MVD平均 ( 7 7± 4 6 )个微血管数 /每平方毫米 ,Ⅲ级和Ⅳ级胶质瘤标本MVD平均 ( 2 3 7± 6 5 )个微血管数 /每平方毫米。Ⅰ级和Ⅱ级胶质瘤与Ⅲ级和Ⅳ级胶质瘤VEGF、VEGFmRNA、MVD之间有非常显著性差异。VEGF和VEGFmRNA与MVD呈正相关。 结论 VEGF与胶质瘤的血管生成密切相关。
Objective To investigate the expression and clinical significance of vascular endothelial factor (VEGF) and angiogenesis in gliomas. Methods The expression of VEGF and VEGFmRNA as well as MVD of low grade (25 cases) and high grade (20 cases) gliomas were detected. Results Over expression of VEGF was fouod in 3 cases of low grade tumors, 18 cases of high grade ones; Lower expression of VEGF was in 22 cases of low grade tumors, and 2 cases of high grade ones. Over expression of proliferating cell nuclear antigen (PCNA) was detected in 2 cases of low grade tumors, and 17 cases of high grade ones; Lower expression of PCNA was 23 cases of low grade tumors, and 3 cases of high grade tumors. MVD was 7.7±4.6 in low grade cases, and 23.7±6.5 in high grade cases. It was significantly different among the expression of VEGF and VEGFmRNA and MVD. VEGFmRNA and VEGF were positively correlated with MVD. Conclusion Expression of VEGF is highly correlated with angiogenesis in glioma, which may serve as a parameter for evaluation of biological behavior, recurrence potential and prognosis of the tumor.
